PEWARNAAN SEL DARAH DENGAN EKSTRAK ANTOSIANIN BUAH NAGA (Hylocereus polyrhizus)

Abstract
Staining of peripheral blood smear assesses morphology of erythrocytes, leukocytes, thrombocytes. Blood smear staining using giemsa synthetic stain. The content in giemsa is not easily decomposed and can generate hazardous and flammable waste. Natural stain can be a non-toxic, biodegradable and renewable stain option. The use of dragon fruit can be an alternative for blood smear coloring based on red color of dragon fruit indicating presence of anthocyanin pigments which can be used as natural stains. The purpose of this research is to observe shape of blood cells as a substitute for giemsa stain. Making dragon fruit extract using infusion method in the form of heating on a hot plate with a temperature of 50°C within 30 minutes. The result of this research was that staining with dragon fruit extract from incubation results showed that erythrocyte cells were well stained and there were also some leukocyte cells. Meanwhile, staining of blood smears with dried dragon fruit extract showed images of erythrocyte cells but not stained properly. The conclusion of this research is that proper drying method to obtain optimal anthocyanin extract can be pursued by drying water content of dragon fruit with a drying method with a stable temperature of 50°C with a duration of time that can adjust thickness of dragon fruit pieces which can range more than 24 hours and waiting time for staining can exceed 45 minutes so that blood cells can more optimally absorb stains from natural stains so that blood cells can be stained properly.
